RT PCR animation - This lecture explains about the RT PCR also known as the real time PCR. Realtime PCR is a technique of amplifying DNA fragments with polymerase chain reaction and along with the PCR. it can help us to monitor the concentration of amplified DNA Replication in real time with the help of fluorescence emission. Real time PCR or RT PCR uses fluorescence resonance energy transfer or Fret to detect the fluorescence generated from the DNA amplification.

I think you've mixed up the nomenclature a bit. The video is about is qrtPCR (or qPCR) - quantitative real time polymerase chain reaction. RT-PCR stands for reverse transcription PCR, which is a different method from qrt-PCR, involving no fluorescent dyes as RNA is electrophoresed and Southern blotted :)

Heading is rt- PCR. rt means reverse transcriptase mediated PCR. But the matter is based on Real Time PCR or qPCR

I'm a med student and rt-PCR it's a pretty used acronym which stands also for Real-Time PCR or qPCR too. I think there is big scientific misunderstanding.

so it seems like your quantitation is dependent on having excess amounts of probe. How do you account for nonspecific DNA binding due to having excess probe?

I have read that flourescence is shown when the probe paired with the dna acc to u it shows after the degradation of probe?

In real-time PCR, Why Extension occurs just after annealing?
Why the primers/probe are added first not the dnTP's. What makes dna polymerase not working then?

Does Dna Polymerase always need s a primer(sequence) to start it's job?

This is not RT-PCR. You are teaching quantitative-PCR (qPCR).
Kindly change the name of the technique.
