The Sanger Method of DNA Sequencing

This is a short animation detailing the steps involved in the original Sanger Method of DNA Sequencing. I hope you enjoy :) Also please note a mistake I made, DNA synthesis occurs in 5' to 3' direction!!

When a 3 minute video describes a topic better than a college professor lollll

DNA polymerase synthesizes in the 5'-3' direction, meaning it reads the template strand in the 3'-5' direction. It needs a 3'OH to extend from. In this animation you place the primer on the 5' end which is not right

Last 15 seconds really made it click for me regarding how the heck the bases are translated from lines to letter sequences. Thanks!

*Note for others watching*
DNA/RNA is synthesized 5'->3' end and template is read 3'->5'. so the primer would have needed to been placed on the 3' end of the template strand. (not the 5' end suggested in the video)
One way I remember this is "5 to 3, I build me" (me and three rhyme.)
"3 to 5, reading is high" (high five)
Hope that helps anyone!

You cut down an 8 minute lecture on this topic to 3 minutes. With great visuals. And music. I love you.

These short videos are more informational and time saving than school and college classes

This video has an error. The synthesis of DNA occurs in 5' --> 3´direction. The primer should be design to anneal in the 3' extremity of DNA template strand.

Nicely explained… but damn i cant believe how much humanity has evolved and advanced.. This is so difficult to even understand and learn i cant imagine how Sanger must’ve invented this … And overcoming the practical challenges in performing the experiment is a whole another challenge

Thank you for this lovely animation. Now I finally understand the Sanger Method better.

2 errors.
1. Denaturation is by NaOH, heat is primarily used in chemical method of sequencing.
2. Primer binds to the 3' end of the DNA

I find it absolutely shocking that you created this excellent animation using only Powerpoint.
Amazing, thank you.

Best video over Sanger's Method of Sequencing I have seen thus far

The Primer is not on the good side at 0:32 and the DNApol goes in the wrong direction 1:20

Very good but the primer has to be attached to the 3 prime end of the template strand.

The DNA polymerase in this video is not synthesizing in the right direction, as the video has it polymerizing in the 3' to 5' direction, which isn't correct. The primer should be attaching to the 3' end of the template strand, and polymerase should be acting in the 5' to 3' direction instead.
