How to plate spore dilutions for single spore (haploid) isolation. Once they germinate, monokaryotic (hopefully) colonies will be checked for the absence of clamp connections and subcultured. They'll then be clear for use in mating experiments.

Got a part 2? How do you know for sure you only got a single spore? I suppose you need a microscope, or when it grows out can you distinctly tell. I know you did a big serial dillution, just trying to better understand how breeding is done and you look to know a few things for sure.

Reverse osmosis water ok? Should I adjust the PH? My RO is ph5.9 with a TDS of 0 .
