Spores dilution

6. Fill the coring bottle (orange cap-rectangular bottle) up to the 200mL mark with DEPC-Treated water (Purple cap bottle).
7. Set the pipette to 200uL, attach on the tip, and draw 200uL spores suspension from the cup in step 5.
8. dispense the 200uL into the orange cap- rectangular bottle.
9. repeat steps 7 and 8 Nine more times, for a total of 10 times.
10. shake and invert the orange cap-rectangular bottle and dispense them to individual 15mL snap-cap tubes.

11. Once the orange cap-rectangular bottle is empty (you do not need to clean/rinse it), add 200mL of DEPC-Treated water.
12. Repeat steps 7 through 9. ( For this batch of 200mL, do not dispense them to individual snap-cap 15mL tubes. Instead, keep them in the orange coring bottle and keep it on ice. Dispense them in the field when you need some. (Remember to invert/shake prior to use/dispense.)
