Breeding Colorado Oyster Mushroom - Pairing Single Spore Isolate Haploids (Immaculate inoculation)

This is the third week into our 2022 breeding project and the single spore isolates are ready for pairing! Hi Gary, thank you so mush for sharing these videos. Your mushroom knowledge has been an amazing source of education for me in my pursuit to cultivate mushrooms. I am wondering if it is possible to breed two strains using complete (not from spore-just two clean and colonized agar wedsges) mycelial cultures, rather than by using 2 haploid cultures from spores to create a diploid? Just curious, as I am inspired to breed some of my strains after seeing your videos, but haven't grown from spore yet. Thanks again for your time. Love what you do man.

Hi Gary, really love the technical content, please keep up the great work! I have always grown from multispore inoculation and was wondering if monokaryonic mycelium looks the same as dikaryonic mycelium? Is there a way to tell (other than trying to fruit it) if a culture is a mated pair?
