-Four types of ELISA
direct ELISA
Indirect ELISA
Competitive ELISA
Sandwich ELISA

The procedure will vary for every test parameter and manufacturer

The procedure to be followed should be based on the manufacturer's instruction and should be validated before being put in use for the patient sample

In here , Ag is coated on to wells by passive adsorption.

Then we add patient's serum sample which contains Ab and it will bind with Ag

Finally enzyme labeled Ag and substrate are added. Enzyme reaction occurs and colour of the solution will change

Allow the reagents in kit to reach room temperature

Plate is coated with a suitable capture Ag

Add conjugate reagent in to each wells
Then cover the plate with the plate sealer and incubate the plate for one hour at 37 celsius

After incubation, remove and discard the plate sealer.
wash each wells 5 times with diluted wash buffer.
After the final washing cycle,turn the strips plate on to blotting paper and tap the plate on to blotting paper and tap the plate to remove any remainders

Colouring(substrate)
Dispense chromogen A and chromogen B solution in to each well
cover the plate with plate sealer and mix gently by tapping the plate

Incubate the plate at 37 celsius for 15 minutes, avoiding light
(In here, Enzyme substrate reaction will take place)

Stopping reaction

Finally measure the absorbances using ELISA reader
