SDS PAGE (Polyacrylamide gel electrophoresis) is a common lab technique used for the separation and detection of proteins. SDS PAGE uses polyacrylamide gel that is formed by the polymerization of acrylamide and cross linking agent bis-acrylamide in presence of APS and TEMED. Native Polyacrylamide gel electrophoresis (native PAGE) separates proteins based on the size as well as net electric charge on the protein. Where as in SDS Polyacrylamide gel electrophoresis (SDS PAGE) the proteins attain the net negative charge and the separation occurs based on the size of proteins.

Chloride ions move faster creating reversed local electric field inverted of that of the mail gel system, which slows the SDS-bond protein behind the chloride ions. However, and with time, the glycine accumulates behind the proteins helping to pack them on the same line. Hiving these forces enough time, the final results will be a sharp distinct three layers: Chloride, proteins, then glycine (from bottom to top)

thanks a lot sir, i had a big doubt how chloride ions bring proteins to same line to separate if it moves first . now i understood that glycine is actually moving first and chloride behind is pushing it move further thus bringing proteins to same line

In stacking gel ... 1st cl ion will move then protein then glycine ... Order will be cl, protein, Glycine (From Down to Up)

why does chloride ion comes later than glycine if they both have negative charge initially?
