Binding and Transport of Vitamin D3: Inside the enterocytes, cholecalciferol binds to a vitamin D-binding protein (DBP) and is transported to the endoplasmic reticulum.

Hydroxylation Reactions: In the liver, vitamin D3 is hydroxylated by the enzyme 25-hydroxylase, leading to the formation of 25-hydroxyvitamin D [25(OH)D], the major circulating form used to assess vitamin D status.

After ingestion, vitamin D3 and vitamin D2 are both subjected to a similar absorption process. They pass through the stomach and enter the small intestine, where they are solubilized by bile acids and pancreatic enzymes. The presence of dietary fat is essential for optimal absorption since vitamin D is fat-soluble.
