Biologist here. The brain stem is generally the most humane way to kill something as it doesnt have pain receptors. But if you miss the spot, youre done. Tissues surrounding the brain stem have a lot of pain receptors.

There are many hate comments down there. Let me explain. When you stab that first spike into the fish, the fish is dead at that point. you put an extra wire to cut all nerves in the spine, which will keep fish fresh and tender. Also, when you cook it it won't fall off apart. When you stab the second wire, they move because the wire is stabbing nerves and when nerve is touched, it makes muscle move. So that's why it seems like he is stabbing a fish that is alive, but it is already dead when he stabbed the first spike.

Edit) only Japan and South Korea uses this technique because they usually eat fish as raw. Not only they it as a raw, they also eat as fermented sushi. If you just simply kill it and ferment it for few days, the fish is too dry and the texture is bad. However, if you cut all nerves systems from the spine, even if you ferment them for a while, the texture stay hard.

Ikejime prevents the muscles from building up lactic acid due to suffocation and thrashing, which both "muddies" the flavor and causes the meat to spoil faster- which makes it unsuitable for sashimi. Fishermen who use ikejime do so because it *works*- you want sashimi grade as often as possible on these fish because it can increase the sale value by quite a lot!

Absolutely true on the most humane thing if people are actually wondering. Leaving fish alive on a stringer or in a small pen stresses them out the whole time and slowly suffocates many species, even in water (most species have to keep moving to stay alive). The meat fills up with acids as the fish suffers, just like human muscles filling with lactic acid under stress. As a result, and bonus, quickly killed and bled fish taste much, much better than those left to linger. It’s almost guaranteed the next fish you eat won’t be treated this nicely, respect your hook and line fisherman who takes care of his/her kill humanely.

No ikejime kit is needed. The very same thing can be achieved with a spike or ice pick inserted quickly and directly into the hindbrain, usually located slightly behind and above the eyes, thereby causing immediate brain death and paralyzation. When spiked correctly, the fish fins flare and the fish relaxes, immediately ceasing all motion and preventing ATP which produces lactic acid and ammonia, making the fish sour, mushy, and less tasteful. The blood contained in the fish flesh retracts to the gut cavity which must be bled out by cutting through a main artery located towards the bottom front of the fish in between the gill plate opening, which produces a better-colored and flavored fillet and prolongs shelf life. Once brain-dead, the fish immediately ceases to feel anything. This method is considered to be the fastest and most humane method of killing fish.
