6:04 If my calculations are correct, if you have a viable cell count of 200, 00 cells/mL in a total volume of 20 mL, then that should be 4, 000, 000 cells. If you then want to to seed 10, 000 cells into the new flask (0.0025 or 0.25% of the total cells), you should transfer 0.05 mL (50 mcL) of the original sample into the new flask.

Wow, counting cell with inverter microscop. Please mention the type of hemocyometer ?

6.07/6:05: Formula= Desired number of cells to seed/Total number of cells=Volume to transfer/Volume of the cell suspension

to transfer/20mL

which is equal to 50 microliter
Further
Total and total volume is 20000ul so 1ul contains= which multiply by 50= 50*200=10000 cells

For anybody stressing over the calculations at the end, these are wrong.
