Because of the resveratrol scandal I have now replaced all my resveratol supplements with flourosent dies ! Works great and as an added bounus I am now a walking light-source without batteries !!😂

After studying a lot of pharmacology, physiology, anatomy, I have a lot of hesitation in taking any supplement / medication. I try to limit the amount as much as possible, because there’s a very fine balance in the body, there’s immense intelligence in the body and the way it maintains homeostasis. Usually overloading your body with an unusual amount of some mineral, vitamin, or compound is often a highway to creating some benefit in some areas, but problems in others. And this is where people begin to add on more and more things to counteract the effects of the things they are taking..
So far, I only take vitamin d3 during winter months alongside vitamin k2 (if I’m not eating enough fermented foods). I feel ok taking these because I know for certain I don’t get enough from my environment or my diet. But even then I have caution, I think people should educate themselves on the complexities of the body because you can make much more informed decisions that way.

I have been taking resveratrol for more than a year and yesterday I was looking for videos on YouTube to send to a friend of mine to convince him to do the same, when I stumbled with your channel and I am very thankful that I did! Few months ago I was diagnosed with high cholesterol and my doctor prescribed me medicine, however, last week my tests didn't show any improvement, despite the fact that I have an active health style, eat very healthy, and maintain a reasonable weight. I think I have identified the culprit and I will stop taking it right away! Can't thank you enough.

Resveratrol is still a senolytic molecule, so it doesn't matter if it activates sirt1 or not (even though dozens of studies and reviews you omitted show it does). It clears out senescent cells and is effective over 18 years of research.

Totally forgotten by now are the two brothers who were doctors in Canada, the Dr. Shute brothers who treated around 30, 000 heart patients with (they felt) much success over many decades. They wrote some books. Don't know if they published any results in the medical journals. However, their clinical experience should not necessarily be discounted because of lack of publication. Sometimes lack of publication merely means lack of funding to do the studies. So the issue is not exactly settled. In any case, this occurred decades ago and is long forgotten.

The only downside is that my girl-friend complains about insomnia !

A good analysis and review. I don't take vitamin E anymore (I never took large amounts knowing that alpha was only one form of the vitamin) but I do have issues with several of the studies on vitamin E. I remember reading an excellent article a long time ago (in "Scientific American Medicine" I think) explaining how vitamin E is part of a cycle : Vit. E -> Selenium -> Vit. C (I don't remember the exact details but those details are not important here for the discussion).
In the article they explained how vit. E once it is oxydized can be harmful and it has to be restored/recycled in order to avoid becoming harmful. They explained how vitamin C and selenium were needed for that. Learning this, it was clear to me that we should not take vitamin E alone especially in large amounts and studies should take that into account and always include a group with vit. C and/or selenium.
Unfortunately, a lot of the studies did not include vit. C and/or selenium. I do remember seeing some studies where there was a vit. E and selenium group and that group had good results compared to the vit. E alone group. However, it would be nice to have a meta-analysis looking more carefully at studies with vit. E combined with at least vit. C and/or selenium. I realize that this is more complicated but that is simply something that has to be dealt with.
I also have issues with the argument that taking a small amount of antioxidant (200ui of alpha-tocoperol, or 500mg of vitamin C) pushes the balance between oxidant and antioxidant to a point where there is too much antioxidant. There are so many antioxidant naturally present in the body
Glutathione concentration in the liver is around 6, 400 micromol per kilogram and Uric acid is around 1600 micromol (wikipedia article on antioxidant - with good references)
Uric acid in human serum is around 300 micromol per kilogram
You would need to take a lot of vitamin E to get a serum or liver concentration that would actually crowd out uric acid
What I do is I make sure I do not take large amounts of one specific nutrient that would not reflect what is found in food so I take a multivitamin.
However, I definitely think there is something that we do not understand and that more research is needed.

Yes resveratrol does not directly active Sirt1 but it does have some activity at activating the Nrf2 system. I have been following resveratrol since the late 1990s.

I’ve been taking metformin for over 10 years and it has helped lower my weight and my A1C (note that I am not nor have ever been diabetic). So as Matt Kaeberlein recently said in an interview: If you’re insulin resistant (which normally means you are overweight too) metformin is helpful. If you’re totally healthy metabolically then you should skip it. But the reality is that most people today are metabolically unhealthy.

Vitamin E is a crucial supplement in the modern world due to the vastly higher intake of polyunsaturated fats in my opinion. Vitamin E requirement goes up in this situation. Its chain-breaking antioxidant capacity is quite unique in this sense. The Shute brothers used very high doses to treat heart disease on thousands of patients. I think a lot of the fear-mongering on vitamin E comes from poor-quality research on synthetic forms of the vitamin which portrayed it in a bad light. I appreciate you're waiting for a stronger safety profile but I do feel that if you look at the breadth of the research, the safety concerns are not as warranted as you might think.
