You know after watching this scene so many times I think it's meant to reflect on some level how cold kosem has become. In this scene she's more angry that Safiye has tricked her into killing her own sister rather than the fact that she's killed he own sister. It's only in two later scenes, the first where she goes through Yasmin's stuff and remembers the past. The second where she's moping in her room about to let go and mourn that Ahmed comes into her room and he rests in her arms. In both these scenes she mourns her sister but more than that I think she realises how far she has gone and the awful things she is capable of doing to keep ahmed safe. Internally she realises and is horrified by the fact that she doesn't really feel that bad about it which she tells Ahemd without directly telling him the details.
