The way Murray treats Arthur reminds me a lot of getting bullied in school. That feeling of you can't say anything right, the bully will find a way to mock what you're saying and the crowd will cheer for it, even if it's not even that clever.

The combination of the trombone and that little old lady lecturing the Joker like he's her grandson sticking his hand into a cookie jar is priceless.

This joke was pivotal to the whole scene. The joker made the sick joke to expose the hypocrisy in society. The audience hated the joke but they are part of a society which throws mentally ill people onto the sidewalk like trash. He even laughed telling it to make it more sick so audience would react

I still laugh at this joke. When I saw this at the cinema, everyone in the audience laughed at this scene.

I feel like his knock-knock joke means more than just a random dull joke to add to the movie's tension. There is a scene where Joker imagines Murray saying that he would love to have a son like him or something like that. That was when he was relatively sane and stable. There is another scene where he finally accepts his inner-self, dresses up as the Joker we know, dances around, then gets chased by cops. During the chase, he literally gets hit by a car. Maybe he was trying to tell Murray that the son he had(at least in his imagination), a peaceful person who did not hate the world around him was now dead, and that all that is left of him is the unstable Joker.

"I ain't got nothing left to lose. Nothing can hurt me anymore."

-That line was honestly heartbreaking. Arthur was hanging by a thread in terms of his mental health at the beginning, but he was an overall good-hearted man who cared for the people around him. But then the world shamelessly tore him apart to the extent that all his empathy disappeared.

The actress sitting next to him does a very nice job. While everyone else is laughing, she's either smiling uncomfortably or not smiling at all--clearly conveying, "I can tell there's something seriously wrong with this guy and I'm in easy reach of him if he snaps."
