He WAS probably reading most of the script for the first time. John Mulaney wrote the scripts for Stephon, and Bill Hader would perform them brilliantly. But between the late afternoon dress rehearsal and the late-night LIVE broadcast, Mulaney would always insert one or two things that Hader had never seen before. And they would always make Hader crack.

It's because John would always give him an entirely new script the day of the show completely throwing Hader off, which is why he is constantly breaking character

This is such poor entertainment reporting. It’s well known that mulaney would change up the Stefon script just before air, so it’s not speculative at all to say Hader was reading many jokes for the very first time live on air

That last statement at the end is pretty much true. The lines for Stefon are written ahead of time with changes made before airing live to catch him off guard and cause him to break character on live tv. That’s been known about for awhile.

He's laughing because of John's voice. In rehearsals, Shy would talk more like Stefon, but on the night John spoke in his normal voice and Bill didn't expect it
