Three different possibilities:
1. Medusa was assaulted by Poseidon and Athena cursed her as punishment for defiling her temple.
2. Medusa was assaulted by Poseidon and Athena blessed her by making is so no one could harm her again (kinda, since she ends up murdered anyway).
3. Medusa and Poseidon slept together and Athena cursed her as punishment for defiling her temple.
The "fourth" possiblity is implausible since Athena wouldn't bless her if Medusa did willing sleep with Poseidon.
