I found your channel while studying for EPPP, and the material presented is amazing. Love the resources provided on your website to follow along as well, which is a great visual & learning tool too!

Nice job. One suggestion - Hyperalgesia from opioid withdrawal/ tolerance is now considered to be mostly driven by glial activation. Opioids activate toll-like receptor 4, and after they stop blocking pain receptors, the neighboring glia remain activated and continue to dump glutamate, substance P, etc. See Linda Watkins' work from U Colorado.

What I don't understand is how would this is considered an adequate defense mechanism. If the person needs to take more and more of the drug to get the same desired effect, Isn't the brain effectively causing the receptors to get damaged? The tolerance + 2 pendant seems like it would cause the person to do more harm to themselves, Rather than protect them.

I know it's not really scientific to ask why, And this is more a philosophical one. But I'm just curious.

can we down-regulate our pain receptors by putting ourselves in pain (like torture)? basically increasing our pain tolerance.

Does tolerance decrease when a person abstaines?

forgive my simple mind but in theory if we use a drug that has the same effect of painkillers but sort of like, happiness killer..?
a drug that lowers the amount of dopamine, wouldn't that cause the receptors to up-regulate, building a tolerance for depression - sadness in the process?

Why can't we simply just make a drug that increases receptors in brain for dopamine and serotonin instead of increasing the amount of serotonin and dopamine transmitters?

Why would you do it in this format? I have the patience of a 21st century ADD patient. I can't handle how long it takes you to make a point, I'm surprised you have so many subscribers. I need more patience, and if you want to blow up big time you have to edit it down so the points are quickly made... it kills me u don't make points efficiently, still appreciate what you're doing but holy gosh.
