No, Reed did not use it to destroy a part of Galactus. He only threatened to use it.
And here's a fact about the Ultimate Nullifier: It not only completely (ultimately) destroy the target, it also destroys whoever uses it.
So Galactus can't be partially destroyed by it, and if Reed had used it, Reed would have been destroyed, too.

He is not only vulnerable to attacks that target his nind, but also attacks that target his soul, as well. When Galactus was at one of his weakest points ever, he returned to Earth to consume our planet. He was so weak, his stature was only that which could be achieved by Giant Man. Even so, it took the combined might of both the FF and the Avengers to slow him down enough for Dr. Strange to hit him with a soul-searing spell that made the big purple guy feel the weight of all the souls he'd extinguished throughout the eons (it wasn't stated, but it's my theory Strange's spell was similar to Ghost Rider's Penance Stare in effect). Galactus collapsed into a coma-like state.
There were many who wanted to kill the world eater so that he could no longer be a threat. But Reed argued against it, pointing out that Galactus was woven into the cosmic fabric of existence and was thus an integral part of the universe, and killing him would have horrible repercussion for the universe, or some such cosmic gibberish (this story took place way back in the early 80s, and it's been a long time since I read that particular book). So the assembled heroes somehow infused Galactus with enough energy that he could leave Earth and find some other world to consume.
Reed would later have to defend himself and Earth at a trial put on by the survivors of the worlds that Galactus had eaten (they traveled in a HUGE armada of spaceships - think Battlestar Galactica's fleet times 10, 000) in the "The Trial of Reed Richards" storyline.
