Translational Silencing by miRNA

Steps Explained:
1. RNA Pol II and III - A miRNA is transcribed into pri-miRNA that folds into a hairpin structure
2. Cropping- Nuclear microprocessor complex crops pri-miRNA(hairpin loop) into pre-miRNA which is exported to nucleus by Exportin 5
3. In cytosol DICER cleaves pre-miRNA (hairpin loop) into miRNA.
4. Incorporation of mi RNA into RISC(RNA inducing silencing complex).RISC are endonucleases called Argonaute proteins which are needed for miRNA induced silencing and contain 2 conserved RNA binding domain :

(i) PAZ Domain (bind to single stranded- 3' end of mature miRNA)
(ii) PIWI Domain (structurally resembles Ribonuclease H)
