You left out one of the best ones: Doom never uses his pinkies when holding something, implying that they're fake since toons only have four fingers.

Honestly, the biggest hint towards Doom being a Toon is his general behavior. His clothes, his attitude, his hammy way of speaking, the fact that he invented a substance that could outright kill Toons in a horrific manner, and the fact that his name is literally Doom all make it clear from the moment he shows up that he's pure evil. No sane person would act so blatantly evil and suspicious... but a Toon would.

I disagree with the first one. Those chemicals in DIP are still very caustic and nasty. A regular human would want to avoid them as well.

I honestly did NOT notice the wind effects on Doom until now. That his villainous Toon nature still affected him while in disguise was genius!

I think "he wouldn't know unless he was a toon" is actually a moot point, because in a world where humans and "real" toons live and work together, most toon things should be well known to humans.

Who Framed Roger Rabbit? is a master class in how to execute a plot twist. There are clues throughout that seem obvious in retrospect, but they're subtle enough that most people wouldn't pick up on them.

Two of my favorite fun/interesting facts about Who Framed Roger Rabbit are:

1) Charles Fleischer, who voiced Roger, also would stand in for him either during blocking or off camera during an actual take and would do his lines in Roger's voice all while dressed like Roger Rabbit with baggie red overalls and bunny ears. As bizzare as this is Bob Hoskins said it really helped him in his performance as Valiant.

2) The plot for WFRR is recycled from a script called "Cloverleaf" which was to be a second sequel to Chinatown with Jack Nicholson reprising his role as private eye Jake Gittes. However, after the previous movie "The Two Jakes" underperformed at the box office, the project was canceled, and Disney bought the script in order to repurpose it into WFRR. The plot is virtually the same, with a corrupt city official using murder and blackmail as a means to acquire a large piece of real estate that will become much more valuable after the freeway is constructed. It's one of the reasons Who Framed Roger Rabbit has that classic film noir feel.

Edit: For some clarification, I forgot that The Two Jakes came out the year after WFRR. Originally, TTJ was going to come out in 85, but production was shut down for four years, which is when Disney optioned Cloverleaf. The box office failure of The Two Jakes did guarantee that Paramount wasn't going to move forward with a third Gittes movie.

The movie also potentially hints at his true toon identity. In R.K. Maroon's office there is a poster on his wall featuring one character whom never makes an appearance throughout the film: Pistol Packin' Possum. This sinister looking character not only displays the same "burnin' red eyes" Doom has, but is also holding the same exact gun Doom used to shoot Maroon.

(And yes, I'm aware of the Marvel graphic novel that gives Doom an origin as "Baron Von Rotten". As none of the filmmakers were involved in that comic, it shouldn't be counted as canon.)

One that you missed was near Doom's introduction. Eddy was picking up a joy buzzer that dropped from Mr. Acme's body. When Doom demanded it back, Eddy used it on Doom. instead of acting like how a normal person would react, Doom's whole body seized up as if electrocuted. the same reaction a toon would have.

I actually have to poke a hole in this one. The reason Doom knew they could laugh themselves to death is because it’s what their “idiot hyena cousins” did. Which Doom knew about.

One thing you notice about Doom is how he tries hard to act as serious as possible but once in awhile his toon persona comes out making him seem uncanny and the impression that something wasn’t right about him

The first time Doom meets Eddie and says "I think you of all people would appreciate that." How would Doom know that Eddie has a hatred for Toons after he lost his brother to a Toon it hints that they had already met before. Even Eddie looks puzzled by this remark he makes.

Did you know the Doom’s plot is actually based on “supposedly” true events. LA had a thriving transit system. The streets cars were magic. Could get you anywhere and you’d never have to drive again while in the city. But a certain car company didn’t like that. Eventually bought up the streets cars, retired them, and helped fund… dun dun dunnnn… freeways. Now people HAD to buy cars. Now LA is one of the most congested cities in North America.

Doom knows the weasels will die of laughter because, as he said, their “cousins” the hyenas died the same way already.

I recently rewatched the show and noticed the awesome foreshadowing in the line about Doom buying the election. I could swear there was one more line that contributes to this but can’t remember.

If only a toon could come up with Doom's convoluted plan, then toons must be real...because the part about buying and scrapping the rail line specifically to promote the creation and use of freeways, and boost private automotive sales, actually happened in the real world.

One really big hint I didn’t hear you mention is that if you look at his teeth, they’re BRIGHT white. Impossibly white for a human. But perfectly white like a toon from the 40s.

With regards to the unblinking eyes, that was Christopher Lloyd's take on the character, and brought it up with the director. Lloyd basically taught himself to not blink between takes.
Also in a game we find out what his true identity is . . . Baron Von Rotten, a common villain in Betty Boop shorts.
