If a drug was released immediately after the Vault was sealed how are the dwellers still alive?

Went to the vault thought it was a glitch because I saw several amatas and two dads

Hey since you’re doing Vaults, can you go over what all these experiments were for?
Like what’s to point of all this, some of these experiments suck or are not that useful when you think about it. This could be like after you cover the vaults
