Here's the crux of the story
1. Ashwathama(Amitabh): At the end of Dwapara Yug(one Yug/era out of 4) Krishna tells him that he will incarnate again as Kalki in Kalyuga(which is our current era) when evil is taken over the world and people are suffering which is depicted all over in the movie. Ashwathama keeps waiting for that day. The moment he sees Deepika, he immediately understands that the child in her belly is the Kalki. His role is to protect her and the child under any cost which is why he tells Deepika that he will protect her.

2.The world: As per Hindu history Kaashi is the first city that came into existence and will be the last before the new era(SatyaYug) begins, which is why everything is centred around that place. That inverted pyramid as per trailer looks like a place where all resources have been stored, and it also looks like its terraforming the earth, meaning those people could be from another planet taking over Earth(or maybe not). The people living outside it are struggling for everything like food and water and everyone is eyeing to get into it but are powerless. So to bring back justice to the world Kalki coming into this wold is important .

3.Prabhas: He's not Kalki(who will ultimately fight with Kaali) as it seems like in the trailer. He is a bounty hunter who's dreaming of getting into that pyramid by collecting 1M units(which is sort of entry fees) somehow. It also seems like Kaal(Kamal Hassan) knows that the child in Deepika's womb is Kalki and is trying to do something with her(like stopping birth or taking control over it or something else) which is why a huge bounty has been declared on her. For Bhairava, this is a golden chance, so he jumps on to capture her and submit her to the army. As Ashwathama is protecting her, both will see a huge fight which you see at the end. Something happens at the end of the fight btw two.
There could be three scenarios for Prabhas here.
a. He himself is the Kalki but isn't aware of his purpose in the world so he is serving as a mere bounty hunter until he meets Deepika and Ashwathama which may help him remember who he is at the end of the fight. Deepika's pregnancy could just be a distraction for the audience.

b. He is a part of Lord Shiva. As per Hindu history, whenever Lord Vishnu has incarnated to serve justice, Lord Shiva(not an incarnation but a part of him) also came into Earth in some form to help him accomplish the task. For example, in Ramayana, it was Hanuman. So Bhairava(which in Sanskrit is also a name of Lord Shiva) is Lord Shiva who is there bcz he needs to help Kalki who will be born later through Deepika to defeat evil. He just doesn't know this. So after fighting with Ashwathama, he may realize that and join hands with Ashwathama to protect Deepika and her child. If this is true we will see more people(meaning more movies) of different entities like Parashuram, Vibhishana, Hanuman, Kripacharya, Vyasa etc(who are supposed to be present in Kalyug just like Ashwathama waiting like him for Kalki) until Kalki is born and grown into adult after which an epic battle will take place between Kaal and Kalki. Bhairav as Lord Shiva will fulfill his destiny by helping Kalki in this case.

c. Again same as scenario a but in this case the possibility is he time travelled to the time when his mother was pregnant for whatsoever reason. But due to time dilation or whatever something must have happened and he might have forgotten who he is but he may recall it when something happens in next part of the movie which is another story about someone like Hanuman or Parashuram or King Mahabali or whoever supposed to help Kalki
