This is quite literally the BEST explanation video of ALS mechanisms that I've ever seen. As a PhD student studying for my qualifying exam, THANK YOU!!!

This is exactly what I was looking for. Everything else is so broad

Thank you - as others have commented, you nailed it. I have been madly doom scrolling everything accessible regarding ALS on the web, and my ChatGPT research has been crazy helpful. With a lot of research only since the beginning of September, I reached out to and was admitted to the Johns Hopkins ALS Clinic. I literally do not know at this moment what’s going on, but they admitted me. I believe the fact that my mother succumbed to ALS at 85, and one of my first cousins was just recently diagnosed. I have mess of symptoms and medical history, which I can trace to everything but ALS. Yet, I go in on Monday to find out what the diagnosis is. I couldn’t be more prepared to advocate for myself, to articulate a solid history of my medical history, with records… not that I am going to beat it, but I sure will meet it. I have hope that somehow this isn’t ALS. Will come back when I know more. Thank you all.

Are the mechanisms of 1) glutamate-mediated excitoxicity and 2) mutant protein aggregation to prions both happening in ALS, or is it one or the another that causes 3) inflammation/release of damaging chemicals?

After watching several videos, I FINALLY get it. Thank you!

This is a best video I watched cleared my understandings of ALS. How could I contact you for more help and guidance.

So that diet the neuropathologist made when she was diagnosed to MS would it not be an effective treatment here? The mylon sheathes were her focus on repairing through diet and she was successful. Why wouldn’t it be successful here around the body instead of the brain.

Thank you so much for this explanation finally i understand the pathology ❤🔥

Trying to like… no option. You are awesome.

it would be interesting to listen more about the genetics of ALS as there are many genes involved in the disease, concepts of mendelian, oligogenic, penetrance...

Very simple but elaborate explanation of the pathophysiology. I am a surgeon myself and is a patient of ALS. I found it to be very useful.

explained in fantastic manner. Thank you very muc.

I don't suppose you have rough estimates of percentages of apoptosis vs. ferroptosis cell deaths. I haven't seen any such numbers yet in published studies. Also curious about the noxious chemicals- are these cytokines, reactive oxygen species, reactive nitrogen species, or something else?

Sadly there are important parts missing in this picture, especially the toxic effects of dietary sugar/fructose on brain metabolism and the effects of insulin resistance on the neurotrasmitter-balance...

It was good and useful. But I think you had a tiny mistake in distinguishing upper motor neurons from lower motor neurons. You said "lower motor neurons in brainstem and spinal cord", while all neurons in these areas are upper motor neurons.
Thanks though, it was pretty useful and clear❤

So, does eating SOD help cure ALS?
Also, why does the gene suddenly mutate at 50 or 60 years of age?

Very excellent explanation. Gives me clear Idea of ALS. Thanks a lot.

Q. Typically how long can it take from discovery of ALS to failure in death ?
