So 20 men and 10 woman could make for an experiment, but what where they doing to need a panther involved?

They probably made it so there were few defense items and plenty of dark places for the panther to hide and maybe even an environment of sorts that the panther can have the advantage and made this Vault as primitive as possible. They probably even picked people with as little survival knowledge as possible to make it authentic, Because Vault-Tec. Panthers aren't even that large but they are known to be powerhouses carrying their prey into trees with only their jaws and their prey can be as large as a water buffalo. Also a Panther can be the family order of the Feline kingdom so a panther could be a Lion or Tiger for all we know. The all black coat is a mutation also that's recessive in African Leopards and Dominant in South American Jaguars.
