Klenow Fragment lecture - The Klenow fragment is a large protein fragment produced when DNA polymerase I from E. Coli is enzymatically cleaved with the aid of the protease subtilisin. First stated in 1970, it retains the 5' → 3' polymerase exercise and the three’ → 5’ exonuclease exercise for removal of precoding nucleotides and proofreading, but loses its 5' → 3' exonuclease activity.

Just a small correction: Exo klenow fragment retains the 5' to 3' polymerase activity not the 3' to 5' exonuclease activity. Mutations can be introduced into the genes of the klenow frament to retain only polymerase activity and no exonuclease activity which is termed the exo klenow fragment. :)

basically the klenow fragment has the same activity as DNA Pol III, so why not using DNA Pol III in the first place?
