Klenow fragment is the part of DNA polymerase I. DNA Polymerase I having 5➡️3 Polymerization and Exonuclease proofreading activity and it also contains 3➡️5 exonuclease activity. If we treat enzyme with Subtilisin protease enzyme then it breaks into two fragments in which one is large and another is small. large fragments having the property of 5➡️3 Polymerization and 3➡️5 exonucleases proofreading activity. This is known as the Klenow fragment. Another fragment having the 5➡️3 Exonuclease proofreading activity but loss the property of Klenow fragment, i.e., it don't have the 5➡️3 Polymerization and 3➡️5 exonucleases proofreading activity
