Tryptophan operon, commonly known as trp operon is regulated at two levels. One is by the trp repressor that inhibits transcription of trp genes when the concentration of tryptophan is high and other is by the leader sequence that forms a termination loop to halt RNA polymerase, causing transcription attenuation.

so basically when trp is low the ribosome is waiting for more trp to come... the wait causes 2 and 3 to form a weak loop and this prevents the strong hair pin between sequence 3 and 4 so no termination occurs.

The trp used by the ribosomes ... that's why they were slowing down when the trp is low .. that's make so much sense thanks professor!!!

How exactly does the loop structure "unbinds" the RNA polymerase? I cannot imagine how this structure interrupts the transcription. You mentioned weak A-U bonds, which break easily. But they are also present when the loop structure is formed between Seq2 and Seq3.

But overall: great video, I now understand (most) of the trp-Operon (:

What happen to poly U region when sequence 4 remains free? How can we say that it will continue the process of transcription while U region is exposed??

So after the Formation of enough Trp, the transcription termination loop halts the Polymerase.
But what about the repressor? Is the repressor just active at higher concentrations or does it work simultaneously?
By the way, briliant video!
