If your Lexus with the 1mzfe with the 4 speed automatic transmission and had both the speed sensors and knock sensors replaced, if the car still wont go into overdrive you will need to drop the transmission and replace the overdrive senser that is inside the transmission, its job is to see how hot the transmission fluid is and then will allow the car to shift into overdrive
Yes i have been fighting this problem on my es300 and hopefully this will resolve the issue. Hope this can shed light on people who have gone crazy with this 4 speed automatic and if i remember correctly almost every Lexus with selectable overdrive will have a overdrive senser
The Transmission Fluid Temperature sensor measures the temperature of the transmission oil (AFT). Its primary function is to inhibit overdrive and converter clutch operation when cold.

If anyone is having this issue with Highlander, but you know you have overdrive initially when you start the car but deactivates 10 or 15 minutes into your drive and you know that this is a knock sensor or ABS light related, I have found a workaround and in the meantime if you drive with the ECT snow button engaged, you will maintain overdrive function for some reason. It’s a little slow to take off from dead stop but at least you’ll have your fourth gear for now either works because it’s keeping you out of the first gear and therefore no high revs to set off knock sensors. I read somewhere that these Toyotas only look for knock sensor readings upward upwards of 3200 RPM or something so if you can manage to keep it low you might preserve your fourth gear function
