Hey Sarah, Biotin is a cofactor, but the name of the enzyme is biotin carboxylase, because the enzyme is dependent on biotin. So it can be called biotin carboxylase, or biotin dependent carboxylase.

biotin carboxylase transcarboxylase is called actecyl-CoA carboxylase (ACC), the other names are the names of it different components. Also the steps of condensation, keto reduction, dehydration and enoil redution are performed by the fatty acid synthase I, and the enlargement of the chain by 2 carbons happens always in the FAS I, by consecutive cycles til they reach 16 Carbons, ACP is a domain of the FAS I, which through a phosphopantethein arm binds the acetil group and malonyl group and the fatty acids in formation through out the reaction

The enzyme for the condensation reaction is beta-ketoacyl (ACP) snthase not synthetase, as no ATP is used. Thought I should mention it in case anyone was confused. Great vdo btw!

Hi there! I wanted to point out something that has been bothering me a bit about these videos. In the introduction of these videos you draw two intertwined snakes around a pole as representation of medicine (or doctors in medical field) I am assuming? I wanted to point out that the symbol is incorrect and in fact, the symbol of medicine and health is rod of asclepius (sp?) which is a single rod with only 1 snake around it. It's more than just a "little difference" since the staff of hermes stands for profit/commerce and even theft (which is what you draw and many people incorrectly assume it be so), whereas, the rod stands for healing as it was carried by Asclepius (Greek god of healing). Just a fyi. :)

It's beta-leto acyl ACP synthase instead of synthatase isn't it. Synthatase would require a nucleotide triphosphate energy input.

6:43 Short fatty acid can become long through addition of hydrocarbons with enzyme fatty acid synthatase???i think this point was not so clear..I would like to try to make it clear.
Actually fatty acid synthase is a multi enzyme complex and refers to all the enzyme responsible for this process. So when you started with Acetyl CoA and Malonyl CoA (with 2 carbons basically) you get after the one complete round a fatty acid with 4 carbons ..your end product of first round would look like (for example R-CO-CH2-CH2-CH3).. now to get longer fatty acid chain you cant just add hydrocarbons rather you have to go one complete round starting from Malonyl CoA and get 2 carbon more at the end . SO for example to produce Stearic acid at the end which has 16 carbons you have to make 8 rounds...during this 8 rounds you spent 1 Acetyl CoA, 8 Malonyl CoA and 16 NADPH + H+.

i don't know if this makes sense to other viewers.

beta keto acyl acp has an extra carbonyl group which isn't supposed to be there. It is still there in hydroxyacyl ACP, Still there in trans 2 enoyl ACP, still there in trans enoyl ACP and here you also forgot to add the carbon carbon double bond> If it says trans it must have a carbon carbon double bond somewhere.

there is a mistakein the four step reaction.while going dehidration and after reduction again you've lost one O molecule.normally reduction can only break the double bonds but in here the double bond has released the O molecule

The structures of B-ketoacyl ACP and B hydroxacy ACP are wrong the should have one less CO2 because it was lost during B-ketoacyl synthase reaction.

thanks very helpful .. but the acetyl coa doosnt bind to ACP it binds to SH group of cysteine of beta -ceto acyl synthase which is in the same enzyme complex (fatty acid synthase) as ACP
