Whenever there is an impending danger, Peter senses it with his spider sense. But here he did not sense anything and became surprised at 2:41 because there was actually no danger but an illusion created by Quentin Beck. That is an extremely wonderful detail as the only danger Peter cannot sense is that of Venom's attacks due to the klyntar symbiote sharing his arachnoid DNA and here it shows that Peter did not sense it as there was no danger.
