The difference is in the sound when you make the marks. John's sounds like how it would in the first game.
Fun Fact: Also if you can notice in game, John closes one eye when he aims while Arthur keeps both open. Which an experienced gunslinger would know helps you aim better for depth. Showing how Arthur was more experienced than John. Although John does keep both open in the first game set 4 years after the epilogue of RDR2

I don't really see any differences but I can tell you this, if your dead eye core becomes empty/turns red your hand will shake ingame. Also, activating your dead eye will automatically reload your gun.

I do think there are differences. I like how Arthur’s one is loud and powerful and it’s a nice way of showing his experience with using weapons and ammunition, he has been the lead enforcer of the gang for quite some time so it sounding like that makes sense. Compared to John’s which is much more of a quiet but deadly deadeye as if he is some kind of ninja. Both very cool.

I once heard that your honor affects it as well. High honor will have the sound of heart beats in dead eye while low honor will be silent or the likes.
Don't have the game to test it tho.

Arthur's clock sound means his not long time in this world. John's heartbeat sound means that he lives, fights and dies for his family

Wow! I genuinely never noticed this difference in RDR2. I always knew John had a different Red Eye in RDR1, but he also has this same one in The RDR2 Prologue. Arthur has a more crisp sound when he marks a target and the hue seems to be lighter compared to John’s. Really cool detail

The sound effect of painting markers on targets is different as well as the overall back ground sounds. John's being quieter and "matter of fact", like his personality. While Arthur's is loud and bold. Another difference is when their Dead Eye is running out. When John's runs low its as if hes being sucked out of the focus. Where as, Arthur's dead eye running out sounds like a clock ticking down. Could be a symbol of his time running out due to his terminal illness.

John had heartbeat noise because he was high honor but arthur had none cause he was low honor, the creator did this btw

Arthurs DE is clock ticking and has somewhat a aggressive paint?

John's DE is a heartbeat the paint doesn't sound aggressive (to me)

Reloading while in DE shows some differences to.

The colors are different. John's Dead Eye is a bright yellow tint while Arthur's is more sepia. Obviously their shooting styles are different.

You gotta think if this is John when you barely start playing as him his dead eye is no where compared to Arthur’s at all, gotta level it up I believe

To those who can’t spot the differences

When activating dead eye, As John it is the same sound when activated in the first game game

When marking, Arthur has a ticking sound while john’s marking sounds are those of the first game

As John, the white streak that changes the screen filter moves left to right, and as Arthur it movies right to left

That’s all I could spot
