A genome-scale CROP-seq screen reveals mediators of T cell signaling for target discovery

In a 10x Genomics webinar, Dr. Tilmann Buerckstuemmer, co-founder and CSO at Myllia Biotechnology, describes the use of genome-wide CRISPR screens in target discovery for immuno-oncology. This CRISPRi screen targeted over 18,000 genes with approx. 80,000 sgRNAs, close to 1 million Jurkat cells were retrieved and analyzed for their single-cell transcriptome. Both known and new regulators of TCR signaling have been recovered, and we aim to translate similar CROP-Seq screens to primary human immune cells.
