I like to think that the laughter is a coping mechanism for the characters. People tend to resort to dark humor and rueful laughter when faced with awful situations, and you can't get much more awful than being trapped in Lordran.

Every one of these laughs conveys a different emotion. Even out of context, the tone of each laugh gives it a different meaning. That's the mark of good voice acting right there.

When you're trapped in Lordran with the world ending and your sanity fading, sharing a laugh or two with someone is probably the most comforting feeling. If I was an NPC myself I'd laugh my ass off to anyone who's not trying to kill me at first glance.
