Southern blotting is a crucial technique in molecular biology used to detect specific DNA molecules in a sample. In the first step, DNA molecules from the sample are separated on a gel using Gel Electrophoresis. Next, the separated DNA fragments are transferred to a Nylon membrane. This transfer involves a special arrangement.

First, the Agarose gel is placed on a sponge which is dipped in a buffer. Now, the Nylon membrane is placed on the gel. Next. we place the filter paper, a glass plate and weight above the Glass Plate.
