The reason dio didnt use his vampire powers is becuase he was so enfatuated with the world and so egotistical he never bothered to use his vampire powers, focusing entirely on the world.

Pretty sure I recall Araki confirming that DIO still had all of his vampiric abilities but the World was so OP that he didn't rely on them nearly as much as he could have
