Why can't we track the cell dynamics for example the vesicular transport with a fluorescent antibody. GFP and antibodies all attach to the target protein and by following their fluorescence pathway, we will be able to observe their movement. Can you please make this point clearer?

Hello, I study a bachelor biology and I don't understand something. If we put the GFP sequence instead of the gene body, how does the gene get transcribed? without a gene body? and only with a promotor?
