In this lecture we are going to discuss “Membrane Attack Complex (MAC)”

Membrane Attack Complex (MAC):
Collection of complement component proteins that are inserted into the cell membranes of micro-organisms and punch holes that result in lysis of the micro-organisms.

The complement components of the MAC are C5b, C6, C7, C8, and multiple copies of C9.

Formation of MAC:

Step:1
First step in the formation of MAC is generation of C5b.
This C5b is formed as end result of the three initiation pathways on pathogen membrane.

Step:2
C5b binds C6 and C7, the resulting complex undergoes a conformational change as a result C7 component inserted into the interior of the microbial membrane.

Step:3
C8 component is attached to C5b67 complex and is inserted into the interior of the phospholipid membrane.

Step:4
Final step in the formation of the MAC is the binding and polymerization of C9 to the C5b678 complex. As many as 10 to 19 molecules of C9 can be bound and polymerized by a single C5b678 complex.This complex generates hole in membrane of pathogen.

MAC-Induced Cell Death:
Formation of these holes in the cell membrane facilitate
1. Free movement of small molecules and ions.
2. Calcium influx into the cytoplasm.
3. Nuclear fragmentation.

cell membranes unable to maintain osmotic integrity, and
the cells lysed after massive influx of water from the extracellular fluid.
