When Odin says : "Only one of you can ascend to the throne, but both of you were born to be kings", what he means is that he plans for Thor to take the throne of Asgard and place Loki on the throne of Jotunheim as a puppet-king controlled by Asgard. It's a cold and heartless strategy. You can also see Loki pick up on the double-meaning and stare at Odin with a confused look... Thor is oblivious, of course.
