The great thing about this scene is the fact that Leonard initiated the kiss. Usually in Hollywood we see the underdog getting kissed by his long time crush. But no, this scene is real because Leonard is the guy who obviously had greater feelings towards Penny. And to kiss her after she said, "Let's talk" suggests that he didn't want her to second guess things. He wanted her to know that he can take charge in a relationship.
