Here restriction enzyme has already been used to cut the dna molecule at specific sites and the small dna fragments needed to be cut out can be separated from the dna molecule by the use of physics. Electric field is applied to the agrose gel containing plate and the dna is inserted in it. The smaller dna molecules(negative) move towards the anode( postive) at a faster rate due to size difference. Like this the required cut dna segments are extracted first. Use of ethidium bromide and uv light is shown so that dna is visible.Then the cut dna are removed from the gel by the process called as elution and can be used for joining with vector dna ( sticky ends concept) with the help of dna ligase enzyme.
