Tripoli means "three cities" so as Tripoli you should conquer three city that reflect its History. The province of Tarablus to reflect the Phoenician origins of the city. Constantinople for the Greek population and influence. And the last choice should be Rome (for the Roman rule of the city) or Toledo (for the Spanish crown attempts to conquer and keep the city). To spicy it up have "the knights" as your vassal (the knights occupy Tripoli for years) and conquer Malta for them.

For Tripoli :
Since Tripoli means Tri-Polis (three cities), you should conquer all cities with Poli in the name :
Constantinople (constantinopoli)
Edirne (adrianopoli)
Bolu (poli)
Gelibolu (Galipoli)
Tarabulus Al sham (Tripoli in lebanon)
Naples (Neapoli)
Felibe (Filipopoli)
Nigbolu (Nicopoli)
This way it will be challenging, you will have to fight Naples, the Ottomans and the Mamluks, 3 strong nations in the med but not too hard to defeit either (lets ignore the russian cities such as Kargopol, conquering from Lybia to Novgorod would be boring)
