Demonstration - Wet Packing a Chromatography Column

Professor Davis briefly demonstrates his technique for wet packing a chromatography column, explaining the proper use and purpose of each component of the column. It's all about uniform length, width and density!

great demonstration, but there is so much missing information.

First of all, the silica needs to be soaked in the mobile phase for a few minutes with more agitation to ensure it is all wet, especially when dealing with larger amounts of silica. Secondly, after pouring in the silica slurry, the column needs to be tapped continuously until all the settling stopped, after which the column needs to be inspected for cracks and/or bubbles. Also, no need to wait for it to settle to check the level. Based on the initial silica volume, and assuming it is all poured into the column (via washing the beaker), you can calculate the level of silica (pretty easy from volume of cylinder equation). Also, a way of speeding up the settling process is by using compressed air.

I’ve been waiting for the follow up video for 4years, I’m starting to think we might never get it! 😵‍💫

Very helpful video in this time of online classes... But please provide the 2nd part of the video demonstrating sample separation

perfect demonstration, cleared a lot of things up for me! But there's no part 2?

happy new year and Thank you so much for this video. Is there standard to use silica gel in column, I mean if my sample a little bit like (0.01 gram), how much of silica gel use here?
in contrast, if I have a lot of samples such as (1 gram or more), here, how much of silica gel have to use?
I mean is there ratio from silica gel to sample to use in column, please tell me

If I have solid sample containing components,
can I use any solvent to dissolve it?
Or I have to use only the same mobile phase as solvent to dissolved sample?
Another case, if must use same mobile phase, but my sample can not dissolve, here, please I need solution to this problem. can I use any solvent? thank you alot in advance

confused by calling the silica gel 'packed' when compared to squeezing out the dmc it is
simply settled and still relatively 'loose' . valuable info missing by u stopping before show
us the sample-load step and the run-the-column after that.
i'm curious too if the flow rate of the dmc thru the column changes / slows much
with the sample added.
thank goodness many others have posted col' chr' vid's here. will visit and see
what all other ideas / methods are applied to this lab method

Hi, can I use alumina oxide for this packing technique ?
