What happened at the end is that Seek came back to the nest with Grumbles Mom infected, along with the sign she used to train Party Grumble. The grumble you saw in Mischievous Lights realm was Party grumble. Party Grumble would've joined his mom in Seeks possession if it wasn't for Guiding Light defeating Mischievous Light.

With the Seek scene, I think the implication was that the “Seek juice” has some sort of hypnotic effect and Seek was coming to hypnotize a new set of Giggles into Screeches

The Seek infected Grumble at the end is the first queen because you see Seek attack her at the beggining and i belive that when the Grumbles were battling against Seek thats when the Seek Goop started to infect her. Your Welcome XD

The grumble at the and was the first queen grumble. At first one giggle tried to eat plush seek but she showed a sign saying "i dont know u cant eat it" and she has her sign even till now. This made me cry like i have never before. This should get awarded with oscar

i always thought that the grumbles were just annoying at first, but when i saw the similarity between the giggles, grumbles and screeches, turns out that the giggles and grumbles are victims to seek, and this show just emphasized that so well. this makes me feel so bad for the giggles and grumbles now OMG. 😭😭
