In this webinar, Dr. John O’Rourke talks about the importance of early cellular testing for antibody functionality during the antibody discovery process. He highlights the need for miniaturized cell-based assays to conserve material, and to screen sufficient numbers of potential candidates using the iQue® Advanced Flow Cytometry Platform.

This webinar includes discussion on:
1. How the iQue® reagent kits are used to perform high-throughput analysis with the iQue® Advanced Flow Cytometry Platform, to quantify antibody dependent cell cytotoxicity, phagocytosis and antibody internalization.
2. The use of iQue Forecyt® software to easily quantify critical functional antibody attributes and reduce time to answer.

0:16 so in this figure we just show the nice spectral separation allowing us to distinguish between Jurkat, Raji and Ramos cells

0:28 in this figure we show heat maps generated in four sites looking at antibody internalizations between Jurkat, Raji and Ramos cells

0:39 levels of antibody internalization and again the negative control is a beta IgG1 and the data here is just summarized in this table looking at the different isotopes what the expectation is for ADCP with IgG1 being much higher than high gA2 and IgG4 and again the EC 50 values in nanograms per ml for each of the different antibodies as well as the maximal ADCP percentage
