I redesigned MLP characters as VILLAINS! I've been seeing a lot of Equestria Girls redesigns as well recently but my drawings are solely based on the OG series since I've never seen EG. My poison suggestion for twilight: strychnine. It's an anticoagulant used most often in rat poison, though there it's mixed with ultrafine shards of glass to induce internal bleeding. It has a white, powdery texture, and a mildly sweet taste, which would make it easy to disguise- just say it's powdered sugar, or put it in a dissolvable capsule
Anticoagulants are also a useful deterrent for fighting, since any injury you get takes longer to clot, and longer to heal. They can contribute to nosebleeds, causing the fine blood vessels of the nose to burst more easily and with more blood- and, they make for very dramatic head wounds (head wounds already bleed a lot- if you can't clot well, they bleed more)

I am obsessed with the redesigns oh my goodness. And Fluttershy being the mafia boss is actually such a good ideas because as you said no-one would ever suspect someone as kind or as sweet as her. And RARITY. Actually obsessed with her sparkly dress and her fashion style. And twilight looks so unhinged and I love how she’s basically a mad scientist in this. And the expressions I was genuinely scared of Fluttershy and Twilight because they both look so menacing. Great job!!

Why do I feel like Pinkie would own the most popular casino in that town along with lavish parties since she is known to love parties, along with in the show she is friends with everyone, I feel like be in charge of some businesses ran by the mafia, like in public her hair is all poofy and she's all bubbly, but when she's doing a job for fluttershy, her hair would be straight and she'd look like a crazy killer, like her pinkamena look in the show. Maybe to a reference to her main creepypasta she'd prefer to wield a chainsaw, as a character once said 'chainsaws and fun and messy"

Wasn't expecting an explanation on Twilight's hair, but I'm actually really glad to see an artist make an active effort to diversify and be more inclusive with their designs and depictions of characters. I absolutely love your designs so I'd be happy to see what you can come up with for the future. (Maybe with Pinkie Pie)

Also, for someone who didn't really watch the show that much you got a pretty good read on the characters and the stories are actually pretty fitting for the AU. I also love the designs and the nods to their canon counterparts.

My poison suggestion is the hellebore flower as a tea as it was used in ancient time to poison a town through their water sources. Symptoms are Vomiting, Slow heart rate, Difficulty breathing, Diarrhea, Drooling, Lethargy and Weakness. The tea option is so Twilight can serve it to Rarity or have someone that works with her to watch Rarity to give her the tea to drink.

Some symptoms you could give rarity is coughing blood, dizziness, fainting spells, and fatigue (fainting spells would be a funny play on her dramatic fainting in the show)

I like how you made Fluttershy a villain by making The Stare and her occasional moments of smothering intensity her primary personality. There are moments that hint at Fluttershy being more controlling and manipulative than she lets on, so I could definitely see this version working especially if she rationalizes it as keeping "bad people" in line.
