John Rambo would definitely win.
He's a U.S. Army Green Beret who was taught different methods on how to deal with various enemies.

Indeed, John Rambo is exceptionally that.
After five movies, he garnered a total kill count of 490.
Also, he's had access to numerous weaponry in all of the films.
Yet there's one tool that he's had throughout the franchise that also gives him an advantage : his Bowie knife that's been with him since his days in Vietnam. It's even part jungle machete, compared to Roman's camping trip blade.
Furthermore, John Rambo has seen more death and carnage and is aware of how the enemy can gang up on him.
He wouldn't fall for that voice changer crap.
Rather, he take the fight to Roman personally.
And John would be bring with him another iconic piece of equipment that Roman dreads upon first sight : the M60 Machine Gun he used in First Blood and in Part II.
The house that the final confrontation Scream 3 took place in would be reduced to looking like Berlin on V-E Day.
As for Roman, getting bombarded with 7.62mm rounds would turn his flimsy police vest into tattered rags and he perish looking like a tree that got swarmed by one too many woodpeckers.

Roman's odds are zero.
