Smg4 mario it can flip flop between the 2 as saiko has been shown to take Mario hits and dishes them back and other times in relative strength

Karen with her new backstory gives her combat capability and knowledge of killing rather than pure fighting

I don’t think that Karen would win because she is only a side character and she has not been here from the start. That’s first off and second saiko has been here since the very beginning and she knows how to fight better than Karen ever did and I think we can all agree that saiko was one of the strongest girl characters in the show
