True - I just uploaded a slightly altered cover for both paperback and hardback versions of a book that was already published. The process was simple. This is what I did:
1) Prepare your updated cover (I always use PDF/A format for both manuscript and cover, but straight PDF is fine)
2) Open your KDP pages for whatever book you're updating
3) Return to the 'Upload cover' section and upload as normal
4) Amazon then repeats the processing stage and you can then use the viewer to check that your upload is what you expected.
5) Save and then press 'Publish.'
6) Amazon then runs its usual checks and about 36 hours later you get the 'Congratulations' email.
7) Your updated version is then Live.
I'm sure exactly the same procedure applies if you're tweaking the manuscript BUT - it would be essential to not alter the page count or you would then need to create a new cover using a new template.Remember - KDP's bots work to tolerances of a thousandth of a millimetre!
For the above changes to the cover, I didn't alter the edition no. as that didn't seem necessary.
There are a few major flaws in the KDP system (dis I hear someone shout "categories"?), but in essence, it's a brilliant system and process.

I do believe that if you update a typo it does not automatically update on Kindle. So if someone has already downloaded a book that error will not update. Only new books downloaded will be free from that particular typo.
