It's really hard to tell if Tsunade loved Jiraiya or not, but if she did, she should have told that to Jiraiya. Ofc that doesn't mean he wouldn't have gone to fight pain, but still, I think it would have made him happy if he knew that atleast she reciprocated his feelings.
