At 18:22 the actual current is going right to left down the potential gradient (from higher potential at V3 to lower potential at V2). That current is (must be) in the same direction as Is (otherwise KCL would be violated at node V3). Is and R1 are in series and thus must have the same current and in the same direction. The reason I(R1) is listed as negative is because it assumes a current direction from left to right which is opposite the actual current direction.
