Not many people probably know of it but in Granstream Saga, you can get the strongest weapon in the game very early on. before even leaving Arona and the Church. Once you enter the basement take a step back (but don’t go back upstairs) and face the West wall, use the panther eye in your inventory and you will get the Onimaru the most powerful weapon in the game. It will make the entire game very easy so you decide whether to use it or not.

They did the same in Mega Man X2 with Shoryuken. I one-shot killed SIgma when he took his virtual appearance when he appeared in the upper right corner.

The Handcannon in Dead Space 2 sorta makes sense given that Isaac might’ve hallucinated all those necromorphs in Titan in his head when he actually using his overworked imagination.

Honorable mention (though it should have been included) has to go to the real Mega Man buster in Dead Rising. That one 1 shot kills enemies as well as bosses (though sometimes 2 shots with bosses) in the game. It's also possible to unlock it very early in the game (assuming you don't mind repeating a specific route underground that involves running over lots of zombies for a bit) if you managed to get past the 3 escaped convicts in the machine gun armed jeep and get to the underground parking lot on the right from where you exit to encounter said convicts and run over zombies with the 3 vehicles available to you. (1 outside of the underground parking lot, 2 in the underground parking lot.)

The ER7 RFW (or "alien superweapon") on Namalsk (Arma 2: Dayz mod, Namalsk) does huge amounts of blood damage (kills anyone who gets hit), has no bullet travel time, and no bullet drop. It is also rare.
It definitely qualifies on this list :)

Personally, I prefer the PRL on re4 over the Infinite Launcher. It can wipe out enemies even far away. There would be times I'd be running somewhere and find enemies already gone and finding whatever they left behind.

Its a bit harder to get because you need to beat the game on Professional mode, but in the end, its worth it. Bosses don't stand a chance against you. 1 or 2 hits (depending on the boss) and BOOM, fights over.

Strongest hidden weapon in Cyberpunk is also easy to obtain. Early in the game, when you are doing the job in the hotel and the corporation boss enters the story, he leaves his helicopter on the roof. You have to run from your hiding spot, and open it. You will find a very rare pistol and sword. Pistol is average throughout the game despite upgrading, but the katana when upgraded is the best in the game - fast, high damage and a serious critical damage multiplier.
